So now I tried the process explorer and killed all applications which
I thought could be the culprit. So it seems no further application was
running but the hotkeys still don't work.
But I have modified the hotkeys for switching the tabs to strg-left or strg-right an I think this is anyway a better shortcut for switching.
Sad but true.
Can't switch between Tabs

...yes, where only a particular ALT key is damaged...
1. Since you were using the left-side ALT key so far, try the key on the right side.
2. Try ALT+LeftArrow and ALT+RightArrow in the folder pane to check if the ALT key works in the first place. (But first comfirm that the folder pane has some history: A r-click on the pane header should show paths of recently visited folders).

So it boils down to the shortcut eaten up by some other application.
I also felt the need for a keyboard shortcut detector application, but could not find one that really works.
There's the Shortcutkeys from http://www.rjlsoftware.com/software/?shortcutkeys, but it does not list even a single shortcut. (It reports in its status line that my PC has 208 shortcuts, though.)
Nikos/BRX- Any ideas on this line? What I have in mind is a sort of logger that reflects all keystrokes and tells which application caught the last keystrokes and which process got activated...Something like the DBmon but which works on the system in general.
